The focus of the flashback is to the story where the current Fairy Tail arc started: Mavis and Zeref's immortality due to the curse of Anksheram. In the last chapter, we saw Zeref declaring his love for Mavis and in doing so, he also caused her death.
According to the translated spoilers from Manga Helpers (h/t Sir Samuel 016 for the translations), the next chapter picks up when Zeref dumps Mavis lifeless body in front of the Guild.
"Waiting to be resuscitated, her body was sealed inside Magic Crystal by the 2nd Guild master Purehito Hades. Purehito's attempts to resucitate Mavis failed. Purehito noticed the curse of Anksehram, and later on built a tomb of sorts in her birthplace of Heaven Wold Island (Tenroujima)."
There was a brief reference to Yuri and Rita, hinting that they were now together in the Afterlife. This happened 3 years after Mavis died.
Fast forward to 30 years later, the next scene depicts Hades unexpectedly creating Fairy Heart!
"His (Hades) talent and Mavis' immortality fused together to create the permanent magic: Fairy Heart. It can continually emit etherion (maybe hinting at Mavis' astral body projections we've seen)It is unknown how to control it, and from that day was forbidden from use."
Meanwhile, there is a tease about Natsu's right arm being the key to end Zeref's immortality. It is hinted that Alvarez is really after Natsu's right arm as it is a weapon of sorts.
The title of the next chapter (452) will be "Prelude to the Final Battle." Is Fairy Tail ending soon? The way they are setting it up, it looks like all mysteries will be settled and the Final story arc will start soon.
